Vendor description (summary): Boster Bio Anti-COX IV COX4I1 Antibody Picoband® (monoclonal, 4G11) catalog # M05442-1.

Biological background

Biological context: This protein is one of the nuclear-coded polypeptide chains of cytochrome c oxidase, the terminal oxidase in mitochondrial electron transport.

Expression and localization notes: cellular localization: Cytoplasm., tissue context: Highly expressed in liver. Expressed in testis and weakly in colon..

Common research applications

  • Western blotting (WB): Compare COX4I1 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of COX4I1 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify COX4I1-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Cytochrome c oxidase subunit 4 isoform 1, mitochondrial is an enzyme that in humans is encoded by the COX4I1 gene. Cytochrome c oxidase (COX) is the terminal enzyme of the mitochondrial respiratory chain. It is a multi-subunit enzyme complex that couples the transfer of electrons from cytochrome c to molecular oxygen and contributes to a proton electrochemical gradient across the inner mitochondrial membrane. The complex consists of 13 mitochondrial- and nuclear-encoded subunits. The mitochondrially-encoded subunits perform the electron transfer and proton pumping activities. The functions of the nuclear-encoded subunits are unknown but they may play a role in the regulation and assembly of the complex. This gene encodes the nuclear-encoded subunit IV isoform 1 of the human mitochondrial respiratory chain enzyme. It is located at the 3' of the NOC4 (neighbor of COX4) gene in a head-to-head orientation, and shares a promoter with it. Pseudogenes related to this gene are located on chromosomes 13 and 14.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cytoplasm.
  • Tissue details: Highly expressed in liver. Expressed in testis and weakly in colon.
  • Research category: Cell Adhesion Proteins,Epigenetics and Nuclear Signaling,Integrins,Mediator Complex,Neuroscience,Receptors,Transcription
