Product Overview
This rabbit polyclonal antibody detects carboxypeptidase N catalytic chain (gene CPN1) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 52.3 kDa, and the supplier reports an observed band at about 52 kDa.
Carboxypeptidase N catalytic chain (CPN1) is the enzymatic subunit of plasma carboxypeptidase N. It removes C-terminal arginine and lysine from circulating peptides such as bradykinin and complement anaphylatoxins, reducing their vasoactive and inflammatory activity.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat liver tissue, mouse liver tissue, rat C6 cells, mouse HEPA1–6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): Caco-2 cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human CPN1 fragment (Asp158–Asn208), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes carboxypeptidase N catalytic chain as a secreted protein. Tissue expression noted by UniProt (human): Synthesized in the liver and secreted in plasma. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
