Product Overview
This rabbit polyclonal antibody detects G1/S-specific cyclin-D1 (gene CCND1) in human and rat samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 33.7 kDa, and the supplier reports an observed band at about 33 kDa.
Cyclin D1 (CCND1) pairs with CDK4 or CDK6 to phosphorylate the retinoblastoma protein RB1 in G1, releasing E2F transcription factors and allowing cells to enter S phase. Its expression responds to mitogenic signals, and it is frequently amplified or overexpressed in cancers such as breast cancer and mantle cell lymphoma.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): rat testis tissue, human placenta tissue, rat brain tissue, MCF-7 cells, COLO320 cells, SW620 cells, MM231 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with a confirmed signal (flow cytometry): A431 cells.
Immunogen
Synthetic peptide from the N-terminal region of human cyclin D1. It differs from the mouse and rat sequences at two residues.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places G1/S-specific cyclin-D1 in the nucleus, cytoplasm and nuclear membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
