Key elements and design rationale

  • Target: KRT16 (Keratin, type I cytoskeletal 16).
  • Antibody format: Monoclonal; clone EDC-11; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

KRT16 (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Epidermis-specific type I keratin that plays a key role in skin. Acts as a regulator of innate immunity in response to skin barrier breach: required for some inflammatory checkpoint for the skin barrier maintenance. . Reported cellular localization context: Endoplasmic reticulum membrane ; Single-pass type I membrane protein . Endoplasmic reticulum . Melanosome . Identified by mass spectrometry in melanosome fractions from stage I to stage IV (PubMed:12643545, PubMed:17081065). The palmitoylated form preferentially localizes to the perinuclear rough ER (PubMed:22314232). . Tissue expression notes (as provided): Expressed in the hair follicle, nail bed and in mucosal stratified squamous epithelia and, suprabasally, in oral epithelium and palmoplantar epidermis. Also found in luminal cells of sweat and mammary gland ducts.

Research relevance and current trends

  • Research context keywords from the source record include: Class I,Cytoskeleton,Cytoskeleton/ECM,Intermediate Filaments,Signal Transduction.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate KRT16 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect KRT16 expression by Western blot in cell or tissue lysates, Detect KRT16 in FFPE tissue sections by immunohistochemistry, Localize KRT16 by immunofluorescence/immunocytochemistry in cultured cells, Quantify KRT16-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 50 kDa; calculated MW: 51268 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 50 kDa
  • Cellular localization (provided): Endoplasmic reticulum membrane ; Single-pass type I membrane protein . Endoplasmic reticulum . Melanosome . Identified by mass spectrometry in melanosome fractions from stage I to stage IV (PubMed:12643545, PubMed:17081065). The palmitoylated form preferentially localizes to the perinuclear rough ER (PubMed:22314232). .
  • Tissue details (provided): Expressed in the hair follicle, nail bed and in mucosal stratified squamous epithelia and, suprabasally, in oral epithelium and palmoplantar epidermis. Also found in luminal cells of sweat and mammary gland ducts.

We are currently using anti-Cytokeratin 16 Rabbit Monoclonal antibody M03393 for human tissue, and we are well pleased with the ICC results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on monkey tissues as well?
The anti-Cytokeratin 16 Rabbit Monoclonal antibody (M03393) has not been validated for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in monkey you can get your next antibody for free. Please contact me if I can help you with anything.
