Overview

This antibody is intended for detection of KRT19 in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Cytokeratin 19 KRT19 Antibody Picoband® (monoclonal, 3D4) catalog # M02101-2. Tested in IF, IHC, WB applications. This antibody reacts with Human. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Mouse Monoclonal Mouse IgG1
  • Clone number: Clone: 3D4
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human Cytokeratin 19, different from the related mouse and rat sequences by nine amino acids.
  • Molecular weight context: reported MW: 44 kDa; calculated MW: nan
  • Reactivity: Human
  • Applications: IF, IHC, WB

As a monoclonal antibody, the reagent targets a defined epitope, supporting consistency across experiments; epitope masking by PTMs or conformational changes can affect signal.

Biological background

keratin 19. Keratin, type I cytoskeletal 19 is a protein that in humans is encoded by the KRT19 gene. The protein encoded by this gene is a member of the keratin family. It is specifically expressed in the periderm, the transiently superficial layer that envelops the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21. Due to its high sensitivity, KRT19 is the most used marker for the RT-PCR-mediated detection of tumor cells disseminated in lymph nodes, peripheral blood, and bone marrow of breast cancer patients. Keratin 19 is often used together with keratin 8 and keratin 18 to differentiate cells of epithelial origin from hematopoietic cells in tests that enumerate circulating tumor cells in blood. Functional note: Involved in the organization of myofibers. Together with KRT8, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle. Reported localization: Isoform Mitochondrial: Mitochondrion. Expression/tissue context: Expressed in a defined zone of basal keratinocytes in the deep outer root sheath of hair follicles. Also observed in sweat gland and mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, ectocervical epithelium (at protein level). Expressed in epidermal basal cells, in nipple epidermis and a defined region of the hair follicle. Also seen in a subset of vascular wall cells in both the veins and artery of human umbilical cord, and in umbilical cord vascular smooth muscle. Observed in muscle fibers accumulating in the costameres of myoplasm at the sarcolemma in structures that contain dystrophin and spectrin.

Research relevance and current trends

  • Class I: Researchers commonly examine how KRT19 relates to this theme using model systems and orthogonal readouts.
  • Cytoskeleton: Researchers commonly examine how KRT19 relates to this theme using model systems and orthogonal readouts.
  • Cytoskeleton/ECM: Researchers commonly examine how KRT19 relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative KRT19 levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of KRT19 across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
