Biological background

Biological context: Involved in late steps of the endosomal multivesicular bodies (MVB) pathway. Recognizes membrane-associated ESCRT-III assemblies and catalyzes their disassembly, possibly in combination with membrane fission. Redistributes the ESCRT-III components to the cytoplasm for further rounds of MVB sorting. MVBs contain intraluminal vesicles (ILVs) that are generated by invagination and scission from the limiting membrane of the endosome and mostly are delivered to lysosomes enabling degradation of membrane proteins, such as stimulated growth factor receptors, lysosomal enzymes and lipids. In conjunction with the ESCRT machinery also appears to function in topologically equivalent membrane fission events, such as the terminal stages of cytokinesis and enveloped virus budding (HIV-1 and other lentiviruses). VPS4A/B are required for the exosomal release of SDCBP, CD63 and syndecan.

Expression and localization notes: cellular localization: Late endosome membrane. Peripheral membrane protein. Prevacuolar compartment membrane., tissue context: Ubiquitously expressed..

Common research applications

  • Western blotting (WB): Compare CCAR2 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of CCAR2 in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify CCAR2-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: Deleted in bladder cancer protein 1 is a protein that in humans is encoded by the DBC1 gene. Cell cycle and apoptosis regulator protein 2 (CCAR2), previously known as DBC1/KIAA1967, regulates diverse cellular functions including transcription, mRNA splicing, metabolism, and circadian cycle. CCAR2 is a critical regulator of cell death or survival following cellular stresses. CCAR2 is a widely expressed protein involved in the regulation of a variety of transcriptional complexes. High expression of CCAR2 correlates with poor outcomes in many human tumor types such as squamous cell carcinoma (SCC).
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Late endosome membrane. Peripheral membrane protein. Prevacuolar compartment membrane.
  • Tissue details: Ubiquitously expressed.
  • Research category: Cytokines,Immunology,Innate Immunity,Interleukins
