Overview

This antibody is intended for detection of DHODH (Dihydroorotate dehydrogenase (quinone), mitochondrial) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-DHODH Antibody Picoband® catalog # PB10056. Tested in IHC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the N-terminus of human DHODH, different from the related mouse sequence by four amino acids, and from the related rat sequence by two amino acids.
  • Molecular weight context: reported MW: 43 kDa; calculated MW: 42867 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Dihydroorotate dehydrogenase (quinone), mitochondrial; Dihydroorotate dehydrogenase (quinone), mitochondrial. Dihydroorotate dehydrogenase (DHODH) is an enzyme that in humans is encoded by the DHODH gene on chromosome 16. The protein encoded by this gene catalyzes the fourth enzymatic step, the ubiquinone-mediated oxidation of dihydroorotate to orotate, in de novo pyrimidine biosynthesis. This protein is a mitochondrial protein located on the outer surface of the inner mitochondrial membrane. Functional note: Catalyzes the conversion of dihydroorotate to orotate with quinone as electron acceptor. Reported localization: Mitochondrion inner membrane ; Single-pass membrane protein . Expression/tissue context: Expressed in prostate and liver. According to some authors, it is not expressed in brain (PubMed:19094056). According to others, weak levels of expression are measured in some brain locations (PubMed:19359404 and PubMed:18545703). Also expressed in epithelium of the small intestine and large intestine, bile duct, nasal mucosa, kidney, adrenal cortex, epithelium of the gastric mucosa with intestinal metaplasia, gallbladder, intercalated ducts of the pancreas, chief cells of the parathyroid and the corpus luteum of the ovary (at protein level). .

Research relevance and current trends

  • Energy Metabolism: Researchers commonly examine how DHODH (Dihydroorotate dehydrogenase (quinone), mitochondrial) relates to this theme using model systems and orthogonal readouts.
  • Energy Transfer Pathways: Researchers commonly examine how DHODH (Dihydroorotate dehydrogenase (quinone), mitochondrial) relates to this theme using model systems and orthogonal readouts.
  • Metabolic Signaling Pathways: Researchers commonly examine how DHODH (Dihydroorotate dehydrogenase (quinone), mitochondrial) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative DHODH (Dihydroorotate dehydrogenase (quinone), mitochondrial) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of DHODH (Dihydroorotate dehydrogenase (quinone), mitochondrial) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
