Overview

This antibody is intended for detection of EME1 (Crossover junction endonuclease EME1) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-EME1 Antibody Picoband® catalog # PB9804. Tested in Flow Cytometry, IHC, IHC-F, ICC, WB applications. This antibody reacts with Human, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human EME1, different from the related mouse sequence by five amino acids.
  • Molecular weight context: reported MW: 62 kDa; calculated MW: 63252 MW
  • Reactivity: Human,Rat
  • Applications: Flow Cytometry, IHC, IHC-F, ICC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Crossover junction endonuclease EME1; Crossover junction endonuclease EME1. Crossover junction endonuclease EME1 is an enzyme that in humans is encoded by the EME1 gene. It is mapped to 17q21.33. This gene encodes a protein that complexes with methyl methanesulfonate-sensitive UV-sensitive 81 protein to form an endonuclease complex. The encoded protein interacts with specifc DNA structures including nicked Holliday junctions, 3'-flap structures and aberrant replication fork structures. Also, this protein may be involved in repairing DNA damage and in maintaining genomic stability. Alternative splicing results in multiple transcript variants. Functional note: Interacts with MUS81 to form a DNA structure-specific endonuclease with substrate preference for branched DNA structures with a 5'-end at the branch nick. Typical substrates include 3'- flap structures, replication forks and nicked Holliday junctions. May be required in mitosis for the processing of stalled or collapsed replication forks. . Reported localization: Nucleus, nucleolus . Recruited to regions of DNA damage in S-phase cells. Expression/tissue context: Highly expressed in thymus, spleen and bone marrow compared to non-hematopoietic tissues such as small intestine, liver, or kidney tissues. Colocalizes with GSK3B and TAU in the Alzheimer disease (AD) brain. Elevated levels seen in breast and colon carcinomas, and which correlates with tumor progression and invasiveness or risk of progression. .

Research relevance and current trends

  • Cell Cycle: Researchers commonly examine how EME1 (Crossover junction endonuclease EME1) relates to this theme using model systems and orthogonal readouts.
  • Cell Differentiation: Researchers commonly examine how EME1 (Crossover junction endonuclease EME1) relates to this theme using model systems and orthogonal readouts.
  • Developmental Biology: Researchers commonly examine how EME1 (Crossover junction endonuclease EME1) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative EME1 (Crossover junction endonuclease EME1) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of EME1 (Crossover junction endonuclease EME1) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
