Product Overview
This rabbit polyclonal antibody targets FAS-associated death domain protein (gene FADD) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 23.3 kDa, and the supplier reports an observed band at about 28 kDa.
FADD is an adaptor protein with a death domain and a death effector domain. It is recruited to activated death receptors such as FAS and TNFR1, where it brings in caspase-8 or caspase-10 to form the death-inducing signaling complex (DISC). FADD is essential for extrinsic apoptosis and also regulates necroptosis and immune signaling.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
|---|
Samples with a confirmed band (WB): human HT1080 cells, human A549 cells, human Jurkat cells, human A431 cells, human HepG2 cells, human HeLa cells, human MCF-7 cells, human Raji cells, rat spleen tissue, rat PC-12 cells, mouse spleen tissue, mouse kidney tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide from the C-terminal region of human FADD.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places FAS-associated death domain protein in the cytoplasm. Tissue expression noted by UniProt (human): Expressed in a wide variety of tissues, except for peripheral blood mononuclear leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
