Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.
Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: E. coli-derived human FLIP recombinant protein (Position: M1-D376). (reported region: M1-D376).
- Molecular weight context: reported MW: 43 kDa; calculated MW: nan
- Reactivity: Human,Mouse,Rat
- Applications: ELISA, IHC, WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
CASP8 and FADD-like apoptosis regulator. CASP8 and FADD-like apoptosis regulator, also called c-FLIP (FLICE-like inhibitory protein), is a protein that in humans is encoded by the CFLAR gene. The protein encoded by this gene is a regulator of apoptosis and is structurally similar to caspase-8. However, the encoded protein lacks caspase activity and appears to be itself cleaved into two peptides by caspase-8. Several transcript variants encoding different isoforms have been found for this gene, and partial evidence for several more variants exists. Functional note: Apoptosis regulator protein which may function as a crucial link between cell survival and cell death pathways in mammalian cells. Acts as an inhibitor of TNFRSF6 mediated apoptosis. A proteolytic fragment (p43) is likely retained in the death-inducing signaling complex (DISC) thereby blocking further recruitment and processing of caspase-8 at the complex. Full length and shorter isoforms have been shown either to induce apoptosis or to reduce TNFRSF-triggered apoptosis. Lacks enzymatic (caspase) activity. Expression/tissue context: Widely expressed. Higher expression in skeletal muscle, pancreas, heart, kidney, placenta, and peripheral blood leukocytes. Also detected in diverse cell lines. Isoform 8 is predominantly expressed in testis and skeletal muscle.
Research relevance and current trends
- Cancer: Researchers commonly examine how CFLAR relates to this theme using model systems and orthogonal readouts.
- Caspases: Researchers commonly examine how CFLAR relates to this theme using model systems and orthogonal readouts.
- Cell Death: Researchers commonly examine how CFLAR relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative CFLAR levels across conditions; band patterns may reflect isoforms and processing.
- IHC/IHC-F: assess spatial distribution of CFLAR across tissue regions and cell types using matched controls.
- ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
