Biological background

Protein function (datasheet): May regulate apoptosis, cell proliferation and cell differentiation. Binds beta-galactoside and a wide array of complex carbohydrates. Inhibits CD45 protein phosphatase activity and therefore the dephosphorylation of Lyn kinase. Strong inducer of T-cell apoptosis. .

Scientific background (datasheet): Galectin-1 is a protein that in humans is encoded by the LGALS1 gene. Galectins belong to a family of related beta-galactoside-binding lectins, also referred to as S-type or S-Lac lectins. Its gene is mapped to 22q12-q13. Galectin-1 was expressed in a subset of slowly dividing subventricular zone astrocytes, which included the neural stem cells. It has been considered as an autocrine negative growth factor that regulates cell proliferation.

Cellular localization (datasheet): Secreted, extracellular space, extracellular matrix .

Tissue details (datasheet): Expressed in placenta, maternal decidua and fetal membranes. Within placenta, expressed in trophoblasts, stromal cells, villous endothelium, syncytiotrophoblast apical membrane and villous stroma. Within fetal membranes, expressed in amnion, chorioamniotic mesenchyma and chorion (at protein level). Expressed in cardiac, smooth, and skeletal muscle, neurons, thymus, kidney and hematopoietic cells. .

Sequence similarities (datasheet): Contains 1 galectin domain.

Research relevance and current trends

  • Commonly studied in contexts related to Cell Biology.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.

We ordered your anti-Galectin 1/LGALS1 antibody for IHC on hepatoma last year. I am using human, and We intend to use the antibody for WB next. Our lab want to know about examining hepatoma as well as lung in our next experiment. Do you have any suggestion on which antibody would work the best for WB?
I viewed the website and datasheets of our anti-Galectin 1/LGALS1 antibody and I see that PA1422 has been tested on human in both IHC and WB. Thus PA1422 should work for your application. Our Boster satisfaction guarantee will cover this product for WB in human even if the specific tissue type has not been validated. We do have a comprehensive range of products for WB detection and you can check out our website bosterbio.com to find out more information about them.
I see that the anti-Galectin 1/LGALS1 antibody PA1422 works with WB, what is the protocol used to produce the result images on the product page?
You can find protocols for WB on the "support/technical resources" section of our navigation menu. If you have any further questions, please send an email to support@bosterbio.com
