Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: E.coli-derived human GRP94 recombinant protein (Position: R43-H221). Human GRP94 shares 99.4% and 98.9% amino acid (aa) sequence identity with mouse and rat GRP94, respectively. (reported region: R43-H221).
- Molecular weight context: reported MW: 100 kDa; calculated MW: 92469 MW
- Reactivity: Human,Mouse,Rat
- Applications: Flow Cytometry, IF, IHC, ICC, WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
Endoplasmin; Endoplasmin. Heat shock protein 90kDa beta member 1 (HSP90B1), known as endoplasmin, or GRP94, is a chaperone protein that in humans is encoded by the HSP90B1 gene. It is mapped to chromosome 12q23.3. This gene encodes a member of a family of adenosine triphosphate (ATP)-metabolizing molecular chaperones with roles in stabilizing and folding other proteins. The encoded protein is localized to melanosomes and the endoplasmic reticulum. Expression of this protein is associated with a variety of pathogenic states, including tumor formation. Functional note: Molecular chaperone that functions in the processing and transport of secreted proteins. When associated with CNPY3, required for proper folding of Toll-like receptors (By similarity). Functions in endoplasmic reticulum associated degradation (ERAD). Has ATPase activity. . Reported localization: Endoplasmic reticulum lumen. Melanosome. Identified by mass spectrometry in melanosome fractions from stage I to stage IV. Expression/tissue context: Expressed in heart, skeletal muscle, eye and brain, and at much lower levels in some other tissues. .

- Cancer Metabolism: Researchers commonly examine how HSP90B1 (Endoplasmin) relates to this theme using model systems and orthogonal readouts.
- ER Proteins: Researchers commonly examine how HSP90B1 (Endoplasmin) relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative HSP90B1 (Endoplasmin) levels across conditions; band patterns may reflect isoforms and processing.
- IHC/IHC-F: assess spatial distribution of HSP90B1 (Endoplasmin) across tissue regions and cell types using matched controls.
- IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
- Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins
- Family / similarity context: Contains 1 HMG box DNA-binding domain.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
