Product Overview
Clone Clone: 5E2 is a mouse monoclonal antibody that recognizes ubiquitin C-terminal hydrolase 7 (gene USP7) in human, mouse and rat samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 128.3 kDa, and the supplier reports an observed band at about 140 kDa.
USP7 (HAUSP) is a deubiquitinating enzyme with many substrates, most famously MDM2 and p53. By stabilizing MDM2 it indirectly promotes p53 degradation, while it can also directly protect p53, placing USP7 at the center of the p53 regulatory circuit. It also regulates DNMT1, UHRF1, PTEN and FOXO proteins and is a studied target in cancer research.

Samples with a confirmed band (WB): human placenta tissue, human 22RV1 cells, human K562 cells, human Caco-2 cells, human HepG2 cells, human U-87MG cells, rat testicular tissue, mouse testicular tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human placenta tissue, rat intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells.
Immunogen
Recombinant human HAUSP/USP7 fragment (Leu258–Asp483), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin C-terminal hydrolase 7 in the nucleus, cytoplasm and chromosome. Tissue expression noted by UniProt (human): Expressed in neural progenitor cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
