Vendor description (summary): Boster Bio Anti-HGS Antibody Picoband® (monoclonal, 6C2E1) catalog # M01174-1.
Biological background
Biological context: Plays a role in the mitochondrial apoptosic process. Upon arrival of cell death signals, promotes mitochondrial outer membrane (MOM) permeabilization by oligomerizing to form pores within the MOM. This releases apoptogenic factors into the cytosol, including cytochrome c, promoting the activation of caspase 9 which in turn processes and activates the effector caspases.
Expression and localization notes: cellular localization: Mitochondrion outer membrane. Single-pass membrane protein., tissue context: Expressed in a wide variety of tissues, with highest levels in the heart and skeletal muscle..
Common research applications
- Western blotting (WB): Compare HGS levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of HGS in tissue sections, considering fixation and antigen retrieval effects.
- Flow cytometry: Quantify HGS-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: Hepatocyte growth factor-regulated tyrosine kinase substrate is an enzyme that in humans is encoded by the HGS gene. It is mapped to 17q25.3. The protein encoded by this gene regulates endosomal sorting and plays a critical role in the recycling and degradation of membrane receptors. The encoded protein sorts monoubiquitinated membrane proteins into the multivesicular body, targeting these proteins for lysosome-dependent degradation.
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Mitochondrion outer membrane. Single-pass membrane protein.
- Tissue details: Expressed in a wide variety of tissues, with highest levels in the heart and skeletal muscle.
