Biological background
Biological context: Calcium/calmodulin-dependent protein kinase that functions autonomously after Ca2+/calmodulin-binding and autophosphorylation, and is involved in synaptic plasticity, neurotransmitter release and long-term potentiation. Member of the NMDAR signaling complex in excitatory synapses, it regulates NMDAR-dependent potentiation of the AMPAR and therefore excitatory synaptic transmission. Regulates dendritic spine development. Also regulates the migration of developing neurons. Phosphorylates the transcription factor FOXO3 to activate its transcriptional activity.
Expression and localization notes: cellular localization: Synapse. Postsynaptic density. Dendritic spine. Dendrite., tissue context: High levels found in the brain, heart and lung while lower levels are seen in the liver, kidney and skeletal muscle..
Common research applications
- Western blotting (WB): Compare RAD23A levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of RAD23A in tissue sections, considering fixation and antigen retrieval effects.
- Flow cytometry: Quantify RAD23A-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: UV excision repair protein RAD23 homolog A is a protein that in humans is encoded by the RAD23A gene. The protein encoded by this gene is one of two human homologs of Saccharomyces cerevisiae Rad23, a protein involved in nucleotide excision repair. Proteins in this family have a modular domain structure consisting of an ubiquitin-like domain (UbL), ubiquitin-associated domain 1 (UbA1), XPC-binding domain and UbA2. The protein encoded by this gene plays an important role in nucleotide excision repair and also in delivery of polyubiquitinated proteins to the proteasome. Alternative splicing results in multiple transcript variants encoding multiple isoforms.
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Synapse. Postsynaptic density. Dendritic spine. Dendrite.
- Tissue details: High levels found in the brain, heart and lung while lower levels are seen in the liver, kidney and skeletal muscle.
