Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human HMGN2, which shares 94.3% and 88.6% amino acid (aa) sequence identity with mouse and rat HMGN2, respectively.
- Molecular weight context: reported MW: 18 kDa; calculated MW: nan
- Reactivity: Human
- Applications: Flow Cytometry, IF, IHC, ICC, WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
high mobility group nucleosomal binding domain 2. Non-histone chromosomal protein HMG-17 is a protein that in humans is encoded by the HMGN2 gene. It is mapped to 1p36.11. The protein encoded by this gene binds nucleosomal DNA and is associated with transcriptionally active chromatin. Along with a similar protein, HMGN1, the encoded protein may help maintain an open chromatin configuration around transcribable genes. The protein has also been found to have antimicrobial activity against bacteria, viruses and fungi. Functional note: Binds to the inner side of the nucleosomal DNA thus altering the interaction between the DNA and the histone octamer. May be involved in the process which maintains transcribable genes in a unique chromatin conformation (By similarity). . Reported localization: Nucleus. Expression/tissue context: Expressed at high levels in lung, placenta and thymus and at lower levels in ovary, intestine and brain.
Research relevance and current trends
- Epigenetics and Nuclear Signaling: Researchers commonly examine how HMGN2 relates to this theme using model systems and orthogonal readouts.
- Histones: Researchers commonly examine how HMGN2 relates to this theme using model systems and orthogonal readouts.
Common research applications
- Western blotting: compare relative HMGN2 levels across conditions; band patterns may reflect isoforms and processing.
- IHC/IHC-F: assess spatial distribution of HMGN2 across tissue regions and cell types using matched controls.
- IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
- Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
