Product Overview

Antibody ANC152.2/8H5 reacts with an extracellular epitope of human CTLA-4 expressed on activated T cells. It has been reported to cross react with dog (11, 12), pig, cow and cat CD152.

Human CD152 (CTLA-4) is a cell surface glycoprotein expressed at low levels on activated T cells (1). CD152 is a high affinity receptor for the costimulatory molecules CD80 (B7-1) and CD86 (B7-2) and appears to function as a negative regulator of T cell activation (2, 3).

Immunogen / Antigen

Stimulated human PBL’s and human CD152 Ig fusion protein

Functional Activity

ANC152.2 blocks binding of CD152 (CTLA-4) Ig fusion protein to its CD80/CD86 receptor, and has been reported to modulate suppressive function of human T Regulatory Cells in Vitro (4). ANC152.2 can bind to intracellular CD152 in fixed permeabolized cells. Purified and F(ab’)2 forms of this clone have been used to block proliferation and/or IL-10 production by activated T cells in various models (5, 6, 7, 8, 9) . ANC152.2 blockade upregulated Activation Induced Marker profile in an Acute…

What applications has this antibody been validated for?

This antibody has been validated for FC, EIA, FX, Blocking. Always refer to the product datasheet for recommended dilution ranges and appropriate positive control samples for each application type.

Which species does this antibody react with?

Validated reactivity has been confirmed for Human, Pig, Cow, Cat, Dog. Cross-reactivity with additional species may exist based on sequence homology but has not been confirmed by the manufacturer. For unlisted species, we recommend running a preliminary validation experiment.

What secondary antibody should I pair with this product?

This antibody was raised in Mouse. For indirect detection (WB, IHC, IF), select an anti-Mouse secondary antibody that matches the isotype. For direct detection, check the variant selector for conjugated formats. Confirm secondary antibody compatibility before ordering.

What is the immunogen used to generate this antibody?

Stimulated human PBL’s and human CD152 Ig fusion protein

How should this antibody be stored?

Please refer to the storage conditions on the product label and datasheet. As a general best practice: upon receipt, aliquot into single-use volumes to minimize freeze-thaw cycling, which can progressively reduce antibody performance and lead to aggregation.

Is this antibody suitable for use with my specific cell line or tissue?

Validated reactivity covers Human, Pig, Cow, Cat, Dog samples. Performance can vary across different cell lines and tissue types depending on target antigen expression levels and epitope accessibility. We recommend verifying target expression in your specific model system using a known positive control before proceeding to experimental samples.
