This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.
Biological background
IGHM (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): IgM antibodies play an important role in primary defense mechanisms. They have been shown to be involved in early recognition of external invaders like bacteria and viruses, cellular waste and modified self, as well as in recognition and elimination of precancerous and cancerous lesions. The membrane- bound form is found in the majority of normal B-cells alongside with IgD. Membrane-bound IgM induces the phosphorylation of CD79A and CD79B by the Src family of protein tyrosine kinases. It may cause death of cells by apoptosis. It is also found in soluble form, which represents about 30% of the total serum immunoglobulins where it is found almost exclusively as a homopentamer. After the antigen binds to the B-cell receptor, the secreted form is secreted in large amounts. . Reported cellular localization context: Isoform 1: Secreted. During differentiation, B-lymphocytes switch from expression of membrane- bound IgM to secretion of IgM. Tissue expression notes (as provided): Expressed by all types of trophoblasts as early as 9 weeks of development. Specific for trophoblastic cells except for amniotic epithelium. In adult tissues, the expression is limited to a few epithelial cell types but is found on a variety of carcinoma.
Research relevance and current trends
- Research context keywords from the source record include: RNA Polymerase,Transcription.
- Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
- Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.
Common research applications
- Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
- Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
- Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.
Workflow ideas (metafield): Validate IGHM antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect IGHM expression by Western blot in cell or tissue lysates, Detect IGHM in FFPE tissue sections by immunohistochemistry, Enrich IGHM by immunoprecipitation from lysates for downstream analysis
Notes for experimental interpretation
- Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
- Apparent molecular weight may vary by sample type and processing (observed MW: 80 kDa; calculated MW: 49307 MW).
- Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.
