Product Overview

Clone GDG-9 is a rabbit monoclonal antibody that recognizes interferon regulatory factor 3 (gene IRF3) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 47.2 kDa, and the supplier reports an observed band at about 60 kDa.

Interferon regulatory factor 3 (IRF3) is a transcription factor that drives type I interferon production during viral infection. Sensing pathways such as RIG-I/MAVS, cGAS–STING and TLR3/4–TRIF activate the kinase TBK1, which phosphorylates IRF3 so it can dimerize, enter the nucleus and induce IFN-β and interferon-stimulated genes.

Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry Not specified by the supplier

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human Hodgkin's lymphoma tissue, human astrocytoma tissue, human ovarian cancer tissue, human cervix carcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human IRF3.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places interferon regulatory factor 3 in the cytoplasm, nucleus and mitochondrion. Tissue expression noted by UniProt (human): Expressed constitutively in a variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:1000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry (dilution not specified). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 47.2 kDa; observed band at about 60 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human IRF3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
