Product Overview
This rabbit polyclonal antibody recognizes E3 ubiquitin-protein ligase Itchy homolog in human, monkey, mouse and rat samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 102.8 kDa, and the supplier reports an observed band at about 103 kDa.
ITCH is a HECT-type E3 ubiquitin ligase of the NEDD4 family. It tags many substrates for degradation or altered signaling, including JUNB, c-FLIP and components of TNF and TLR pathways, and it is important for limiting inflammation and preventing autoimmunity, as ITCH-deficient mice develop severe inflammatory disease.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Monkey, Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human K562 cells, human Raji cells, human 293T cells, monkey COS-7 cells, human HepG2 cells, human A549 cells, rat C6 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Synthetic peptide from the middle region of human ITCH/AIP4. It differs from the mouse sequence at one residue.
Reactivity Notes
The supplier lists reactivity with human, monkey, mouse and rat. UniProt places E3 ubiquitin-protein ligase Itchy homolog in the cell membrane, cytoplasm and nucleus. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
