Biological background
Biological context: Acyl-CoA synthetases (ACSL) activates long-chain fatty acids for both synthesis of cellular lipids, and degradation via beta-oxidation. ACSL3 mediates hepatic lipogenesis (By similarity). Preferentially uses myristate, laurate, arachidonate and eicosapentaenoate as substrates (By similarity). Has mainly an anabolic role in energy metabolism. Required for the incorporation of fatty acids into phosphatidylcholine, the major phospholipid located on the surface of VLDL (very low density lipoproteins). .
Expression and localization notes: cellular localization: Mitochondrion outer membrane ; Single-pass type III membrane protein . Peroxisome membrane ; Single-pass type III membrane protein . Microsome membrane ; Single-pass type III membrane protein . Endoplasmic reticulum membrane ; Single-pass type III membrane protein ., tissue context: Expressed in breast, ductal and invasive ductal carcinomas of the breast, sporadic colorectal adenomas and carcinomas (at protein level). Expressed in fetal brain. Expressed in lung, amygdala, eye, prostate, pancreatic and prostate cancers, head and neck tumors and embryonal tumor..
Common research applications
- Western blotting (WB): Compare IWS1 levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of IWS1 in tissue sections, considering fixation and antigen retrieval effects.
- Flow cytometry: Quantify IWS1-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: Protein IWS1 homolog also known as interacts with Spt6 (IWS1) is a protein that in humans is encoded by the IWS1 gene. Various steps in gene expression, such as mRNA processing, surveillance, export, and synthesis are coupled to transcription elongation). The C-terminal domain (CTD) of the large subunit of RNA polymerase II plays an important role in the integration of these different steps. IWS1 interacts with Spt6, a CTD-binding transcription elongation factor and H3 chaperone. IWS1 also recruits another CTD-binding protein, HYPB/Setd2 histone methyltransferase, to the RNA polymerase II complex for elongation-coupled H3K36 trimethylation. Thus, IWS1 links Spt6 and HYPB/Setd2 in a large complex and regulates mRNA synthesis and histone methylation at the co-transcriptional level .
- Cross reactivity: No cross-reactivity with other proteins.
