Key elements and design rationale

  • Target: MIF — Macrophage migration inhibitory factor
  • Antibody format: Host: Rabbit; Clonality: Polyclonal; Isotype: Rabbit IgG
  • Species reactivity: Human
  • Molecular weight guidance: Observed: 12 kDa; Calculated: 12476 MW

Specificity note: No cross reactivity with other proteins.

Biological background

Protein function (datasheet): Pro-inflammatory cytokine. Involved in the innate immune response to bacterial pathogens. The expression of MIF at sites of inflammation suggests a role as mediator in regulating the function of macrophages in host defense. Counteracts the anti- inflammatory activity of glucocorticoids. Has phenylpyruvate tautomerase and dopachrome tautomerase activity (in vitro), but the physiological substrate is not known. It is not clear whether the tautomerase activity has any physiological relevance, and whether it is important for cytokine activity. .

Scientific background (datasheet): Macrophage migration inhibitory factor, MIF, is a cytokine released by T-lymphocytes, macrophages, and the pituitary gland that serves to integrate peripheral and central inflammatory responses. MIF gene has 3 exons separated by introns of only 189 and 95 bp, and covers less than 1kb. Localization of the human gene for macrophage migration inhibitory factor (MIF) to chromosome 22q11.2. MIF plays a critical role in inflammatory diseases and atherogenesis.

Cellular localization (datasheet): Secreted. Cytoplasm. Does not have a cleavable signal sequence and is secreted via a specialized, non- classical pathway. Secreted by macrophages upon stimulation by bacterial lipopolysaccharide (LPS), or by M.tuberculosis antigens.

Tissue details (datasheet): Restricted to a subset of activated T-cells. .

Sequence similarities (datasheet): Belongs to the MIF family.

Research relevance and current trends

  • Commonly studied in contexts related to Immunology,Innate Immunity,Macrophage/Inflammation.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
