Vendor description (summary): Boster Bio Anti-MIRO1/RHOT1 Antibody Picoband® catalog # A05928-1.
Biological background
Biological context: Transcriptional regulator that acts as an activator. Promotes beta-catenin transcriptional activity. Plays a role in tumorigenesis. Enhances the neoplastic transforming activity of CTNNB1.
Expression and localization notes: cellular localization: Nucleus., tissue context: Expressed in breast, ductal and invasive ductal carcinomas of the breast, sporadic colorectal adenomas and carcinomas (at protein level). Expressed in fetal brain. Expressed in lung, amygdala, eye, prostate, pancreatic and prostate cancers, head and neck tumors and embryonal tumor..
Common research applications
- Western blotting (WB): Compare RHOT1 levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of RHOT1 in tissue sections, considering fixation and antigen retrieval effects.
- Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
- Flow cytometry: Quantify RHOT1-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: Mitochondrial Rho GTPase 1 (MIRO1) is an enzyme that in humans is encoded by the RHOT1 gene on chromosome 17. The Ras homolog family member T1 (RHOT) is an atypical Rho Ca2+-binding GTPase that localizes to the mitochondria. RHOT1, the related protein RHOT2, the adaptor protein Milton, and the PTEN induced putative kinase 1 (PINK1), form a complex that is involved in axonal transport of mitochondria. Both PINK1 and Parkin target RHOT1 for phosphorylation and degradation, causing the arrest of mitochondrial motility.
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Nucleus.
- Tissue details: Expressed in breast, ductal and invasive ductal carcinomas of the breast, sporadic colorectal adenomas and carcinomas (at protein level). Expressed in fetal brain. Expressed in lung, amygdala, eye, prostate, pancreatic and prostate cancers, head and neck tumors and embryonal tumor.
