Biological background

Protein function (datasheet): Constitutive NADPH oxidase which generates superoxide intracellularly upon formation of a complex with CYBA/p22phox. Regulates signaling cascades probably through phosphatases inhibition. May function as an oxygen sensor regulating the KCNK3/TASK-1 potassium channel and HIF1A activity. May regulate insulin signaling cascade. May play a role in apoptosis, bone resorption and lipolysaccharide-mediated activation of NFKB. .

Scientific background (datasheet): NADPH oxidase 4 is an enzyme that in humans is encoded by the NOX4 gene, and is a member of the NOX family of NADPH oxidases. This gene encodes a member of the NOX-family of enzymes that functions as the catalytic subunit the NADPH oxidase complex. The encoded protein is localized to non-phagocytic cells where it acts as an oxygen sensor and catalyzes the reduction of molecular oxygen to various reactive oxygen species (ROS). The ROS generated by this protein have been implicated in numerous biological functions including signal transduction, cell differentiation and tumor cell growth. A pseudogene has been identified on the other arm of chromosome 11. Alternative splicing results in multiple transcript variants.

Cellular localization (datasheet): Endoplasmic reticulum membrane ; Multi-pass membrane protein . Cell junction, focal adhesion . Cell membrane . May localize to plasma membrane and focal adhesions. .

Tissue details (datasheet): EXpressed in brain, in all layers of the cerebellum, in pyramidal cells of the Ammon horn and in Purkinje cells (at protein level). Expressed in osteoclasts, leukocytes, kidney, liver and lung. .

Sequence similarities (datasheet): Contains 1 FAD-binding FR-type domain.

Research relevance and current trends

  • Commonly studied in contexts related to Atherosclerosis,Cardiovascular,Cell Biology,Drug Metabolism,Heart Disease,Metabolic Signaling Pathways,Metabolism,Oxidative Stress,Pathways and Processes,Redox Metabolism,Signal Transduction,Vascular Inflammation.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.

Our team were content with the WB result of your anti-NADPH oxidase 4/NOX4 antibody. However we have observed positive staining in nephron tubule endoplasmic reticulum membrane using this antibody. Is that expected? Could you tell me where is NOX4 supposed to be expressed?
According to literature, nephron tubule does express NOX4. Generally NOX4 expresses in endoplasmic reticulum membrane. Regarding which tissues have NOX4 expression, here are a few articles citing expression in various tissues: Fetal kidney, Pubmed ID: 11376945 Kidney, Pubmed ID: 10869423, 11032835 Lung, Pubmed ID: 15721269 Ovary, Pubmed ID: 15489334 Pulmonary artery, Pubmed ID: 14702039
