Overview

Anti-Nedd4 Antibody Picoband® is an antibody reagent for detection of Nedd4 (neural precursor cell expressed, developmentally down-regulated 4, E3 ubiquitin protein ligase). Researchers commonly use anti-Nedd4 antibodies to measure relative expression and localization across biological samples, with assay selection guided by the listed applications (WB, IHC, IF, Flow, ELISA).

Boster Bio Anti-Nedd4 Antibody Picoband® catalog # A00984-4. Tested in ELISA, Flow Cytometry, IF, IHC, WB applications. This antibody reacts with Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Target: Nedd4 (neural precursor cell expressed, developmentally down-regulated 4, E3 ubiquitin protein ligase). Alternative names: E3 ubiquitin-protein ligase NEDD4
  • Antibody format: Polyclonal; Rabbit IgG
  • Species context: Host: Rabbit, Reactivity: Mouse,Rat
  • Purification: Immunogen affinity purified.
  • Immunogen: E.coli-derived mouse Nedd4 recombinant protein (Position: A7-L655).
  • Molecular weight context: observed 115 kDa (reported)
  • Provided application(s): WB, IHC, IF, Flow, ELISA

These attributes help contextualize how the antibody is commonly selected (host/clonality/isotype/label) and how signals are interpreted across sample types and assay formats.

Biological background

Function: E3 ubiquitin-protein ligase which accepts ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then ly transfers the ubiquitin to targeted substrates. Specifically ubiquitinates 'Lys-63' in target proteins (PubMed:23644597). Involved in the pathway leading to the degradation of VEGFR-2/KDFR, independently of its ubiquitin-ligase activity. Monoubiquitinates IGF1R at multiple sites, thus leading to receptor internalization and degradation in lysosomes. Ubiquitinates FGFR1, leading to receptor internalization and degradation in lysosomes. Promotes ubiquitination of RAPGEF2. According to PubMed:18562292 the link between NEDD4 and PTEN regulation through polyubiquitination described in PubMed:17218260 is questionable. Involved in ubiquitination of ERBB4 intracellular domain E4ICD. Involved in the budding of many viruses. Part of a signaling complex composed of NEDD4, RAP2A and TNIK which regulates neuronal dendrite extension and arborization during development. Ubiquitinates TNK2 and regulates EGF-induced degradation of EGFR and TNF2. Involved in the ubiquitination of ebola virus VP40 protein and this ubiquitination plays a role in facilitating viral budding. Ubiquitinates BRAT1 and this ubiquitination is enhanced in the presence of NDFIP1 (PubMed:25631046).

Cellular localization: Cytoplasm. Cell membrane; Recruited to the plasma membrane by GRB10. Once complexed with GRB10 and IGF1R, follows IGF1R internalization, remaining associated with early endosomes. Uncouples from IGF1R-containing endosomes before the sorting of the receptor to the lysosomal compartment (By similarity). May be recruited to exosomes by NDFIP1.

Background: E3 ubiquitin-protein ligase NEDD4, also known as neural precursor cell expressed developmentally down-regulated protein 4 (NEDD4), is an enzyme that in humans is encoded by the NEDD4 gene. This gene is the founding member of the NEDD4 family of HECT ubiquitin ligases that function in the ubiquitin proteasome system of protein degradation. The encoded protein contains an N-terminal calcium and phospholipid binding C2 domain followed by multiple tryptophan-rich WW domains and, a C-terminal HECT ubiquitin ligase catalytic domain. It plays critical role in the regulation of a number of membrane receptors, endocytic machinery components and the tumor suppressor PTEN.

Cross reactivity: No cross-reactivity with other proteins.

Research relevance and current trends

  • Quantitative and spatial profiling: expression patterns are increasingly studied across cell states using multiplex imaging and omics-informed validation.
  • Isoforms and post-translational modifications: researchers often evaluate how isoform composition and PTMs can shift apparent molecular weight or localization.
  • Context-aware interpretation: comparative studies commonly include perturbations (stimulation, inhibition, genetic models) to relate target changes to pathway behavior.

Common research applications

  • Western blot (WB): compare relative target abundance and apparent size shifts (e.g., isoforms/PTMs) across conditions.
  • Immunohistochemistry (IHC): assess distribution across tissue compartments and compare staining patterns between groups.
  • Immunofluorescence / ICC: evaluate subcellular localization and co-localization with compartment markers.
  • Flow cytometry: quantify target-positive populations and compare shifts after stimulation or differentiation.

Across these uses, researchers typically interpret changes in signal as relative differences between matched sample groups, considering sample preparation and biological context.

Notes for experimental interpretation

  • Apparent molecular weight can vary due to isoforms, proteolysis, glycosylation, phosphorylation, and sample preparation differences.
  • Species reactivity and epitope conservation can influence observed signal patterns, especially in cross-species studies.
  • Control concepts: include appropriate negative controls (e.g., isotype controls where relevant) and, when feasible, genetic or orthogonal controls (KO/KD, peptide competition, or independent assays) to support interpretation.

For antibody reagents, monoclonal antibodies are often chosen for epitope consistency across lots, while polyclonals may recognize multiple epitopes and can show different background characteristics depending on context.
