Product Overview
This rabbit polyclonal antibody targets NPC intracellular cholesterol transporter 1 (gene NPC1) in human, mouse and rat samples and is validated for ELISA and Western blot. The predicted molecular weight is 142.2 kDa, and the supplier reports an observed band at about 170 kDa.
NPC1 is a large lysosomal membrane protein that exports cholesterol from late endosomes and lysosomes. It receives LDL-derived cholesterol from the soluble protein NPC2 and moves it across the lysosomal membrane. Loss of NPC1 causes Niemann–Pick disease type C, and NPC1 also serves as the intracellular receptor for Ebola virus entry.

Samples with a confirmed band (WB): rat pancreas tissue, mouse NIH3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places NPC intracellular cholesterol transporter 1 in the late endosome membrane and lysosome membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
