Product Overview
Clone BGA-14 is a rabbit monoclonal antibody that recognizes neurogenic locus notch homolog protein 1 (gene NOTCH1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 272.5 kDa, and the supplier reports an observed band at about 110 kDa.
NOTCH1 is a single-pass transmembrane receptor activated by Jagged and Delta-like ligands on neighboring cells. Ligand binding triggers cleavage that releases the Notch intracellular domain (NICD), which moves to the nucleus and activates target genes with RBPJ. NOTCH1 controls cell fate in many tissues, including T-cell development, and is frequently mutated in T-cell acute lymphoblastic leukemia.

Samples with a confirmed band (WB): human 293T cells, human K562 cells, human Jurkat cells, rat brain tissue, rat PC-12 cells, mouse brain tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human liver tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

The supplier lists reactivity with human, mouse and rat. UniProt places neurogenic locus notch homolog protein 1 in the cell membrane and late endosome membrane. Tissue expression noted by UniProt (human): In fetal tissues most abundant in spleen, brain stem and lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
