Product Overview
This rabbit polyclonal antibody detects NEDD8 ultimate buster 1 (gene NUB1) in human, monkey, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 70.5 kDa, and the supplier reports an observed band at about 75 kDa.
NUB1 (NEDD8 ultimate buster 1) is a negative regulator of the NEDD8 conjugation pathway. It binds NEDD8, FAT10 (UBD) and proteins carrying these modifiers and delivers them to the proteasome for degradation, so it is studied in protein turnover and ubiquitin-like modifier signaling.

Samples with a confirmed band (WB): human HeLa cells, human Colo320 cells, human HepG2 cells, human K562 cells, human Raji cells, monkey COS-7 cells, human MCF-7 cells, human A549 cells, rat liver tissue, rat PC-12 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human adrenal adenomas tissue, human bladder urothelial carcinoma tissue, human ovarian cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Recombinant human NUB1 fragment (Lys190–Asp543), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, monkey, mouse and rat. UniProt places NEDD8 ultimate buster 1 in the nucleus. Tissue expression noted by UniProt (human): Widely expressed with lowest expression in the pancreas for isoform 1 and in leukocytes, liver, prostate and skeletal muscle for isoform 2. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
