Product Overview
This rabbit polyclonal antibody targets nuclear ubiquitous casein and cyclin-dependent kinase substrate 1 (gene NUCKS1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 27.3 kDa, and the supplier reports an observed band at about 20 kDa.
NUCKS1 is an abundant chromatin-associated nuclear protein that is heavily phosphorylated by casein kinase 2 and cyclin-dependent kinases. It binds DNA and supports homologous recombination repair of DNA breaks, so it is studied in genome maintenance and cell proliferation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Mouse, Rat) |

Samples with a confirmed band (WB): human 293T cells, human SH-SY5Y cells, human MCF-7 cells, human A431 cells, rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human esophageal squamous carcinoma tissue, human liver cancer tissue, human lung cancer tissue, human rectum adenocarcinoma tissue, human thyroid papillary carcinoma tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): rat brain tissue, mouse brain tissue.
Samples with a confirmed signal (flow cytometry): HeLa cells.
Immunogen
Synthetic peptide from the N-terminal region of human NUCKS1. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places nuclear ubiquitous casein and cyclin-dependent kinase substrate 1 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Widely expressed, with highest levels in thyroid gland, prostate and uterus and in fetal liver, thymus and lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
