Vendor description (summary): Boster Bio Anti-P-Cadherin-3 CDH3-Antibody Picoband® (monoclonal, 3C9) catalog # M03353.

Biological background

Biological context: Cadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types.

Expression and localization notes: cellular localization: Cell membrane. Single-pass type I membrane protein., tissue context: Expressed in some normal epithelial tissues and in some carcinoma cell lines..

Common research applications

  • Western blotting (WB): Compare CDH3 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of CDH3 in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify CDH3-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Cadherins, such as CDH3, are integral membrane glycoproteins responsible for calcium-dependent cell-cell adhesion. Cadherin-3 is a protein that in humans is encoded by the CDH3 gene. This gene is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein composed of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. This gene is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. In addition, aberrant expression of this protein is observed in cervical adenocarcinomas. Mutations in this gene have been associated with congential hypotrichosis with juvenile macular dystrophy.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cell membrane. Single-pass type I membrane protein.
  • Tissue details: Expressed in some normal epithelial tissues and in some carcinoma cell lines.
  • Research category: ER Proteins,Immunology,Protein Trafficking,Signal Transduction
