Cellular localization Nucleus. Cytoplasm. Endosome . Nuclear and cytoplasmic in quiescent cells. AKT- or RSK- mediated phosphorylation on Thr-198, binds 14-3-3, translocates to the cytoplasm and promotes cell cycle progression. Mitogen- activated UHMK1 phosphorylation on Ser-10 also results in translocation to the cytoplasm and cell cycle progression. Phosphorylation on Ser-10 facilitates nuclear export. Translocates to the nucleus on phosphorylation of Tyr-88 and Tyr-89. Colocalizes at the endosome with SNX6; this leads to lysosomal degradation (By similarity). .

Rabbit Monoclonal

Product Overview

Clone CGA-3 is a rabbit monoclonal antibody that recognizes cyclin-dependent kinase inhibitor 1B (gene CDKN1B) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 22.1 kDa, and the supplier reports an observed band at about 25 kDa.

p27 (CDKN1B, p27Kip1) is a cyclin-dependent kinase inhibitor that holds cells in G1 by blocking cyclin E-CDK2 and cyclin A-CDK2 activity. Its levels are tightly controlled by phosphorylation and degradation, and it is studied as a regulator of proliferation, quiescence and tumor suppression.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places cyclin-dependent kinase inhibitor 1B in the nucleus, cytoplasm and endosome. Tissue expression noted by UniProt (human): Expressed in kidney (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this p27 KIP1 antibody been tested for IHC on prostate tissue?
A.According to the supplier: no. IHC on prostate tissue has not been tested.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 22.1 kDa; observed band at about 25 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human p27 KIP 1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
