Product Overview
This rabbit polyclonal antibody detects protein disulfide-isomerase (gene P4HB) in human, monkey, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 57.1 kDa, and the supplier reports an observed band at about 57 kDa.
Protein disulfide isomerase (PDI, P4HB) is an abundant endoplasmic reticulum enzyme that forms, breaks and reshuffles disulfide bonds in newly made proteins and can also act as a chaperone. It is the β subunit of collagen prolyl 4-hydroxylase and of the microsomal triglyceride transfer protein, and is studied in protein folding and ER stress.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): T-47D cells.
Samples with a confirmed signal (flow cytometry): U937 cells.
Immunogen
Recombinant human P4HB fragment (Ala146–Asp363), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, monkey, mouse and rat. UniProt places protein disulfide-isomerase in the endoplasmic reticulum, endoplasmic reticulum lumen, melanosome and cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
