Vendor notes: Boster Bio Anti-PERK/EIF2AK3 Antibody Picoband® catalog # A01992-2. Tested in ELISA, Flow Cytometry, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E. coli-derived human PERK recombinant protein (Position: R222-Q334). (reported region: R222-Q334).
  • Molecular weight context: reported MW: 140 kDa; calculated MW: nan
  • Reactivity: Human,Mouse,Rat
  • Applications: ELISA, Flow Cytometry, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

eukaryotic translation initiation factor 2-alpha kinase 3. Eukaryotic translation initiation factor 2-alpha kinase 3, also known as protein kinase R (PKR)-like endoplasmic reticulum kinase (PERK), is an enzyme that in humans is encoded by the EIF2AK3 gene. The protein encoded by this gene phosphorylates the alpha subunit of eukaryotic translation-initiation factor 2, leading to its inactivation, and thus to a rapid reduction of translational initiation and repression of global protein synthesis. This protein is thought to modulate mitochondrial function. It is a type I membrane protein located in the endoplasmic reticulum (ER), where it is induced by ER stress caused by malfolded proteins. Mutations in this gene are associated with Wolcott-Rallison syndrome. Functional note: Metabolic-stress sensing protein kinase that phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (eIF-2-alpha/EIF2S1) on 'Ser-52' during the unfolded protein response (UPR) and in response to low amino acid availability. Converts phosphorylated eIF-2-alpha/EIF2S1 either in a global protein synthesis inhibitor, leading to a reduced overall utilization of amino acids, or to a translation initiation activator of specific mRNAs, such as the transcriptional activator ATF4, and hence allowing ATF4-mediated reprogramming of amino acid biosynthetic gene expression to alleviate nutrient depletion. Serves as a critical effector of unfolded protein response (UPR)- induced G1 growth arrest due to the loss of cyclin-D1 (CCND1). Involved in control of mitochondrial morphology and function. Reported localization: Endoplasmic reticulum membrane; Single-pass type I membrane protein. Expression/tissue context: Ubiquitous. A high level expression is seen in secretory tissues.

Research relevance and current trends

  • Diabetes: Researchers commonly examine how EIF2AK3 relates to this theme using model systems and orthogonal readouts.
  • DNA/RNA: Researchers commonly examine how EIF2AK3 relates to this theme using model systems and orthogonal readouts.
  • Epigenetics and Nuclear Signaling: Researchers commonly examine how EIF2AK3 relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative EIF2AK3 levels across conditions; band patterns may reflect isoforms and processing.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

Our team were well pleased with the WB result of your anti-PERK/EIF2AK3 antibody. However we have been able to see positive staining in nasal cavity mucosa endoplasmic reticulum membrane using this antibody. Is that expected? Could you tell me where is EIF2AK3 supposed to be expressed?
From literature, nasal cavity mucosa does express EIF2AK3. Generally EIF2AK3 expresses in endoplasmic reticulum membrane. Regarding which tissues have EIF2AK3 expression, here are a few articles citing expression in various tissues: Brain, Pubmed ID: 14702039 Brain, and Pancreas, Pubmed ID: 10677345 Cervix carcinoma, Pubmed ID: 20068231 Liver, Pancreas, and Testis, Pubmed ID: 10026192
