Key elements and design rationale
- Antibody format: Rabbit Polyclonal Rabbit IgG
- Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of human Peroxiredoxin 4, different from the related mouse and rat sequences by one amino acid.
- Molecular weight context: reported MW: 31 kDa; calculated MW: 30540 MW
- Reactivity: Human,Mouse,Rat
- Applications: Flow Cytometry, IF, IHC, ICC, WB
As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.
Biological background
Peroxiredoxin-4; Peroxiredoxin-4. PRDX4 (peroxiredoxin 4) is also known as AOE37-2. The protein encoded by this gene is an antioxidant enzyme and belongs to the peroxiredoxin family. Functional analysis showed that PRDX4 protects glutamine synthetase from inactivation. Yeast 2-hybrid, immunoprecipitation, and immunoblot analyses indicated that PRDX4 and PRDX1 are capable of homodimerization and heterodimerization with each other but not with the mitochondrial PRDX3. Gel mobility shift and immunoblot analysis found that PRDX4 depletes NFKB binding activity together with a reduction in the amounts of p50, p65, and phosphorylated IKBA, as well as a reduction in the expression of HIV-1 viral proteins. Expression of PRDX4, alone or with PRDX1, increased the resistance of yeast cells to oxidant-induced toxicity. Jin et al. suggested PRDX4 modulates IKBA phosphorylation in the cytoplasm and thus affects a peroxiredoxin-dependent redox step. Functional note: Probably involved in redox regulation of the cell. Regulates the activation of NF-kappa-B in the cytosol by a modulation of I-kappa-B-alpha phosphorylation. . Reported localization: Cytoplasm . Secreted .
Research relevance and current trends

Common research applications
- Western blotting: compare relative PRDX4 (Peroxiredoxin-4) levels across conditions; band patterns may reflect isoforms and processing.
- IHC/IHC-F: assess spatial distribution of PRDX4 (Peroxiredoxin-4) across tissue regions and cell types using matched controls.
- IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
- Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.
Notes for experimental interpretation
- Specificity notes: No cross reactivity with other proteins.
- Cross-reactivity: No cross-reactivity with other proteins.
- Family / similarity context: Belongs to the AhpC/TSA family.
- Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
- Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
