Biological background

Protein function (datasheet): Required for TCR (T-cell antigen receptor)- and pre-TCR- mediated signaling, both in mature T-cells and during their development. Involved in FCGR3 (low affinity immunoglobulin gamma Fc region receptor III)-mediated signaling in natural killer cells and FCER1 (high affinity immunoglobulin epsilon receptor)-mediated signaling in mast cells. Couples activation of these receptors and their associated kinases with distal intracellular events such as mobilization of intracellular calcium stores, PKC activation, MAPK activation or cytoskeletal reorganization through the recruitment of PLCG1, GRB2, GRAP2, and other signaling molecules. .

Cellular localization (datasheet): Cell membrane ; Single-pass type III membrane protein . Present in lipid rafts.

Tissue details (datasheet): Expressed in thymus, T-cells, NK cells, mast cells and, at lower levels, in spleen. Present in T-cells but not B-cells (at protein level). .

Research relevance and current trends

  • Commonly studied in contexts related to Adapters,Adaptive Immunity,G Protein Signaling,Immunology,Non-CD,Ras Family,Signal Transduction,Signaling Pathway,Small G Proteins,T Cells,Transmembrane.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

We ordered your anti-Phospho-LAT (Y191) Rabbit Monoclonal antibody for IF on liver a few months ago. I am using mouse, and We intend to use the antibody for ICC next. We need examining liver as well as leukemia in our next experiment. Do you have any suggestion on which antibody would work the best for ICC?
I looked at the website and datasheets of our anti-Phospho-LAT (Y191) Rabbit Monoclonal antibody and I see that P01654 has been validated on mouse in both IF and ICC. Thus P01654 should work for your application. Our Boster satisfaction guarantee will cover this product for ICC in mouse even if the specific tissue type has not been validated. We do have a comprehensive range of products for ICC detection and you can check out our website bosterbio.com to find out more information about them.
