Product Overview
This rabbit polyclonal antibody detects protein kinase C eta type (gene PRKCH) in human samples and is validated for flow cytometry, IHC, ICC/IF and Western blot. The predicted molecular weight is 77.8 kDa, and the supplier reports an observed band at about 78 kDa.
PKCη (PRKCH) is a calcium-independent, diacylglycerol-activated novel protein kinase C enriched in epithelial tissues. In keratinocytes it promotes growth arrest and differentiation, and it also helps maintain epithelial tight junctions by phosphorylating occludin, so it is studied in skin biology, barrier function and T-cell signaling.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human) |
|---|---|
| IHC (frozen sections) | 0.5–1 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 0.5–1 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): A431 cells, A549 cells, HeLa cells, SKOV cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with a confirmed signal (flow cytometry): A431 cells, K562 cells, MCF-7 cells.
Immunogen
Recombinant human PKC eta fragment (Arg30–Asp389), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 96.7% and 96.1%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places protein kinase C eta type in the cytoplasm. Tissue expression noted by UniProt (human): Most abundant in lung, less in heart and skin. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
