Product Overview
This rabbit polyclonal antibody targets 5'-3' exonuclease PLD3 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and ELISA. The predicted molecular weight is 54.7 kDa, and the supplier reports an observed band at about 55 kDa.
PLD3 is a lysosomal 5′-to-3′ exonuclease that degrades single-stranded DNA and RNA, despite belonging to the phospholipase D family. By clearing nucleic acids in endolysosomes, it limits activation of the nucleic acid sensors TLR7 and TLR9, and it has also been studied as a genetic factor in Alzheimer's disease and spinocerebellar ataxia.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human appendiceal adenocarcinoma tissue, human liver cancer tissue, human spleen tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Immunogen
Recombinant human PLD3 fragment (Met1–Leu490), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places 5'-3' exonuclease PLD3 in the lysosome lumen, early endosome membrane, late endosome membrane and endosome membrane. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
