Overview
Anti-POLA2 Antibody Picoband® is an antibody for POLA2 detection raised in Rabbit (Polyclonal, Rabbit IgG), with reported reactivity: Human. Commonly used in WB, IHC, Flow Cytometry, ELISA workflows.
Key elements and design rationale
- Target: POLA2 (serpin family A member 4); UniProt: Q14181
- Antibody format: Rabbit, Polyclonal, Rabbit IgG
- Molecular weight: 70 kDa, calculated 82457 MW
- Applications: WB, IHC, Flow Cytometry, ELISA
Vendor description (summary): Boster Bio Anti-POLA2 Antibody Picoband® catalog # A08427-2.
Biological background
Biological context: Inhibits human amidolytic and kininogenase activities of tissue kallikrein. Inhibition is achieved by formation of an equimolar, heat- and SDS-stable complex between the inhibitor and the enzyme, and generation of a small C-terminal fragment of the inhibitor due to cleavage at the reactive site by tissue kallikrein.
Expression and localization notes: cellular localization: Secreted., tissue context: Expressed by the liver and secreted in plasma..
Common research applications
- Western blotting (WB): Compare POLA2 levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of POLA2 in tissue sections, considering fixation and antigen retrieval effects.
- Flow cytometry: Quantify POLA2-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: DNA polymerase alpha subunit 2 is an enzyme that in humans is encoded by the POLA2 gene. POLA2 may play an essential role at the early stage of chromosomal DNA replication by coupling the polymerase alpha/primase complex to the cellular replication machinery (By similarity).
- Cross reactivity: No cross-reactivity with other proteins.
- Cellular localization: Secreted.
- Tissue details: Expressed by the liver and secreted in plasma.
- Research category: Cancer,Cell Biology,Cytoskeleton/ECM,ECM Enzymes,Extracellular Matrix,Invasion/Microenvironment,Metalloprotease,Proteolysis/Ubiquitin,Proteolytic Enzymes,Signal Transduction
