Key elements and design rationale

  • Target: PPT1 (palmitoyl-protein thioesterase 1); UniProt: P50897
  • Antibody format: Mouse, Monoclonal, clone Clone: 10F3, Mouse IgG2b
  • Molecular weight: 34 kDa
  • Applications: WB, IHC, Flow Cytometry, ELISA

Vendor description (summary): Boster Bio Anti-PPT1 Antibody Picoband® (monoclonal, 10F3) catalog # M02690.

Biological background

Biological context: Removes thioester-linked fatty acyl groups such as palmitate from modified cysteine residues in proteins or peptides during lysosomal degradation. Prefers acyl chain lengths of 14 to 18 carbons.

Expression and localization notes: cellular localization: Lysosome. Secreted..

Common research applications

  • Western blotting (WB): Compare PPT1 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of PPT1 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify PPT1-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Palmitoyl-protein thioesterase 1 (PPT-1), also known as palmitoyl-protein hydrolase 1, is an enzyme that in humans is encoded by the PPT1 gene. PPT-1 is a member of the palmitoyl protein thioesterase family. The protein encoded by this gene is a small glycoprotein involved in the catabolism of lipid-modified proteins during lysosomal degradation. The encoded enzyme removes thioester-linked fatty acyl groups such as palmitate from cysteine residues. Defects in this gene are a cause of infantile neuronal ceroid lipofuscinosis 1 (CLN1, or INCL) and neuronal ceroid lipofuscinosis 4 (CLN4). Two transcript variants encoding different isoforms have been found for this gene.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Lysosome. Secreted.
  • Research category: Cancer,Lipid Metabolism,Metabolism,Signal Transduction
