Vendor description (summary): Boster Bio Anti-PROM1/CD133 Antibody Picoband® (monoclonal, 7F13) catalog # M01767-4.

Biological background

Biological context: Seems to act as an adapter protein between membrane proteins and the actin cytoskeleton. In collaboration with CBLC, modulates the rate of RET turnover and may act as regulatory checkpoint that limits the potency of GDNF on neuronal survival. Controls CBLC function, converting it from an inhibitor to a promoter of RET degradation. May play a role in receptor clustering and cytoskeletal polarity in the junction between T- cell and antigen-presenting cell. May anchor the podocyte slit diaphragm to the actin cytoskeleton in renal glomerolus. Also required for cytokinesis.

Expression and localization notes: cellular localization: Cytoplasm, cytoskeleton . Cell projection, ruffle . Colocalizes with F-actin and BCAR1/p130Cas in membrane ruffles. Located at podocyte slit diaphragm between podocyte foot processes (By similarity). During late anaphase and telophase, concentrates in the vicinity of the midzone microtubules and in the midbody in late telophase., tissue context: Widely expressed in fetal and adult tissues..

Common research applications

  • Western blotting (WB): Compare PROM1 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of PROM1 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify PROM1-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: Prominin-1, also known as CD133, is a glycoprotein that in humans is encoded by the PROM1 gene. It is mapped to 4p15.32. Prominin-1 is a member of pentaspan transmembrane glycoproteins (5-transmembrane, 5-TM), which specifically localize to cellular protrusions. This gene encodes a pentaspan transmembrane glycoprotein. The protein localizes to membrane protrusions and is often expressed on adult stem cells, where it is thought to function in maintaining stem cell properties by suppressing differentiation. It has been proposed to act as an organizer of cell membrane topology. Prominin-1 was expressed not only on metastatic colon cancer cells, but also on differentiated colonic epithelium in both adult mice and humans.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cytoplasm, cytoskeleton . Cell projection, ruffle . Colocalizes with F-actin and BCAR1/p130Cas in membrane ruffles. Located at podocyte slit diaphragm between podocyte foot processes (By similarity). During late anaphase and telophase, concentrates in the vicinity of the midzone microtubules and in the midbody in late telophase.
  • Tissue details: Widely expressed in fetal and adult tissues.
