Overview

This antibody is intended for detection of PSMA3 (Proteasome subunit alpha type-3) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Proteasome 20S alpha 3/PSMA3 Antibody Picoband® catalog # PB10089. Tested in Flow Cytometry, IHC, IHC-F, ICC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence in the middle region of human PSMA3, identical to the related mouse and rat sequences.
  • Molecular weight context: reported MW: 28 kDa; calculated MW: 28 kDa
  • Reactivity: Human,Mouse,Rat
  • Applications: Flow Cytometry, IHC, IHC-F, ICC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Proteasome subunit alpha type-3; Proteasome subunit alpha type-3. Proteasome subunit alpha type-3, also known as macropain subunit C8 and proteasome component C8, is a protein that in humans is encoded by the PSMA3 gene. The proteasome is a multicatalytic proteinase complex with a highly ordered ring-shaped 20S core structure. The core structure is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides. This gene encodes a member of the peptidase T1A family, that is a 20S core alpha subunit. Two alternative transcripts encoding different isoforms have been identified. Functional note: The proteasome is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. The proteasome has an ATP-dependent proteolytic activity. Binds to the C-terminus of CDKN1A and thereby mediates its degradation. Negatively regulates the membrane trafficking of the cell-surface thromboxane A2 receptor (TBXA2R) isoform 2. . Reported localization: Cytoplasm. Nucleus.

Research relevance and current trends

  • Proteasome: Researchers commonly examine how PSMA3 (Proteasome subunit alpha type-3) relates to this theme using model systems and orthogonal readouts.
  • Proteasome / Ubiquitin: Researchers commonly examine how PSMA3 (Proteasome subunit alpha type-3) relates to this theme using model systems and orthogonal readouts.
  • Proteolysis/Ubiquitin: Researchers commonly examine how PSMA3 (Proteasome subunit alpha type-3) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative PSMA3 (Proteasome subunit alpha type-3) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of PSMA3 (Proteasome subunit alpha type-3) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
