Product Overview
This rabbit polyclonal antibody recognizes pre-mRNA-processing-splicing factor 8 (gene PRPF8) in human and mouse samples and is validated for Western blot and ELISA. The predicted molecular weight is 273.6 kDa, and the supplier reports an observed band at about 250 kDa.
PRPF8 is a very large, highly conserved protein at the heart of the spliceosome. It forms a scaffold that positions the U5, U6 and U2 snRNAs and the pre-mRNA at the catalytic center of both major and minor spliceosomes, and PRPF8 mutations are a cause of autosomal dominant retinitis pigmentosa.

Samples with a confirmed band (WB): human HeLa cells, human SH-SY5Y cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Recombinant human PRPF8 fragment (Glu433–His2166), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places pre-mRNA-processing-splicing factor 8 in the nucleus and nucleus speckle. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
