Vendor description (summary): Boster Bio Anti-RAB27A Antibody Picoband® (monoclonal, 2F5) catalog # M01608.

Biological background

Biological context: Plays a role in cytotoxic granule exocytosis in lymphocytes. Required for both granule maturation and granule docking and priming at the immunologic synapse.

Expression and localization notes: cellular localization: Lysosome. Late endosome. Membrane. Lipid-anchor. Melanosome., tissue context: Found in all the examined tissues except in brain. Low expression was found in thymus, kidney, muscle and placenta. Detected in melanocytes, and in most tumor cell lines examined. Expressed in cytotoxic T-lymphocytes (CTL) and mast cells..

Common research applications

  • Western blotting (WB): Compare RAB27A levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of RAB27A in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify RAB27A-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Ras-related protein Rab-27A is a protein that in humans is encoded by the RAB27A gene. The protein encoded by this gene belongs to the small GTPase superfamily, Rab family. The protein is membrane-bound and may be involved in protein transport and small GTPase mediated signal transduction. Mutations in this gene are associated with Griscelli syndrome type 2. Alternative splicing occurs at this locus and four transcript variants encoding the same protein have been identified.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Lysosome. Late endosome. Membrane. Lipid-anchor. Melanosome.
  • Tissue details: Found in all the examined tissues except in brain. Low expression was found in thymus, kidney, muscle and placenta. Detected in melanocytes, and in most tumor cell lines examined. Expressed in cytotoxic T-lymphocytes (CTL) and mast cells.
  • Research category: Actin Binding Proteins,Actin, etc.,Cytoskeleton,Cytoskeleton/ECM,Microfilaments,Signal Transduction
