Product Overview
This rabbit polyclonal antibody detects rab3 GTPase-activating protein catalytic subunit (gene RAB3GAP1) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 110.5 kDa, and the supplier reports an observed band at about 130 kDa.
RAB3GAP1 is the catalytic subunit of the Rab3 GTPase-activating complex, which switches off Rab3 proteins by accelerating GTP hydrolysis. Together with RAB3GAP2 it also activates Rab18, and RAB3GAP1 mutations cause Warburg micro syndrome, which affects eye, brain and genital development.

Samples with a confirmed band (WB): human 293T cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human glioblastoma tissue, human breast cancer tissue, human liver cancer tissue, human lung adenocarcinoma tissue, human spleen tissue, human testicular seminoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Siha cells.
Samples with confirmed staining (ICC/IF): human lung cancer tissue, human liver cancer tissue.
Samples with a confirmed signal (flow cytometry): HL-60 cells.
Immunogen
Recombinant human RAB3GAP1 fragment (Asn109–Gln810), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places rab3 GTPase-activating protein catalytic subunit in the cytoplasm, endoplasmic reticulum and Golgi apparatus. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
