Product Overview
This rabbit polyclonal antibody detects DNA repair protein RAD50 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 153.9 kDa, and the supplier reports an observed band at about 154 kDa.
RAD50 is an ATPase of the SMC family that forms the MRN complex with MRE11 and NBS1. MRN detects DNA double-strand breaks, activates ATM, begins end resection for homologous recombination and helps maintain telomeres, so RAD50 is central to studies of DNA damage signaling and repair.

|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human Jurkat cells, human MCF-7 cells, human HeLa cells, human K562 cells, rat thymus tissue, rat C6 cells, mouse thymus tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human testis cancer tissue, human thyroid cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Recombinant human RAD50 fragment (Asn1224–His1312), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places DNA repair protein RAD50 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Expressed at very low level in most tissues, except in testis where it is expressed at higher level. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
