Product Overview
This rabbit polyclonal antibody recognizes V(D)J recombination-activating protein 2 (gene RAG2) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 59.2 kDa, and the supplier reports an observed band at about 59 kDa.
RAG2 is the essential partner of RAG1 in the RAG recombinase that initiates V(D)J recombination in developing B and T cells. Its PHD domain reads histone H3 lysine 4 trimethylation to target accessible gene segments, and loss of RAG1 or RAG2 function causes severe combined immunodeficiency.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
|---|---|
| Western blot | 0.1–0.5 µg/mL (Human) |
Samples with a confirmed band (WB): A549 cells, 22RV1 cells, U2OS cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 40 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse spleen tissue, rat spleen tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the C-terminal region of human RAG2. It differs from the mouse sequence at three residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places V(D)J recombination-activating protein 2 in the nucleus. Tissue expression noted by UniProt (human): Cells of the B- and T-lymphocyte lineages. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
