Product Overview

Clone FIF-18 is a rabbit monoclonal antibody that recognizes proto-oncogene tyrosine-protein kinase receptor Ret in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 124.3 kDa; the supplier reports observed bands at 150, 175 kDa.

RET is a receptor tyrosine kinase activated by GDNF-family ligands (GDNF, neurturin, artemin and persephin), which bind GFRα co-receptors and then engage RET. RET signaling is essential for kidney development and the enteric nervous system. Loss-of-function mutations cause Hirschsprung disease, while activating mutations and fusions drive multiple endocrine neoplasia type 2 and some thyroid and lung cancers.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places proto-oncogene tyrosine-protein kinase receptor Ret in the cell membrane and endosome membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 124.3 kDa; observed bands at about 150 and 175 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human ret.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
