Biological background
Protein function (datasheet): Mediates electroneutral potassium-chloride cotransport. May be activated by cell swelling. May contribute to cell volume homeostasis in single cells.
Scientific background (datasheet): Solute carrier family 12 (potassium/chloride transporters), member 6, also called SLC12A6 or KCC3, is a protein that in humans is encoded by the SLC12A6 gene. This gene is a member of the K-Cl cotransporter (KCC) family. K-Cl cotransporters are integral membrane proteins that lower intracellular chloride concentrations below the electrochemical equilibrium potential. By radiation hybrid and somatic cell hybrid analyses, this gene is mapped to 15q14. The proteins encoded by this gene are activated by cell swelling induced by hypotonic conditions. Mutations in this gene are associated with agenesis of the corpus callosum with peripheral neuropathy.
Cellular localization (datasheet): Basolateral cell membrane ; Multi-pass membrane protein .
Tissue details (datasheet): Highly expressed in heart, brain and kidney. Detected at lower levels in skeletal muscle, placenta, lung and pancreas. Detected in umbilical vein endothelial cells. Isoform 2 is more abundant in kidney. Isoform 5 is testis specific. Expressed in the proximal tubule of the kidney (at protein level). .
Research relevance and current trends
- Commonly studied in contexts related to Neurogenesis,Neurology Process,Neuroscience,Neurotransmission,Potassium Channels,Receptors / Channels.
- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
