Biological background
Protein function (datasheet): Transporter for alanine, serine, cysteine, and threonine. Exhibits sodium dependence.
Scientific background (datasheet): Neutral amino acid transporter A, also called SLC1A4 or ASCT1, is a protein that in humans is encodes by the SLC1A4 gene. The SLC1A4 gene is mapped to chromosome 2p14 based on an alignment of the SLC1A4 sequence with the genomic sequence (GRCh37). ASCT1 does not transport glutamate or aspartate but alanine, serine, cysteine, and threonine instead. It is found that the ASCT1 transporter functions primarily as an amino acid exchanger. Transport is associated with a chloride channel activity that is thermodynamically uncoupled from animo acid transport. This gene also exhibits sodium dependence.
Cellular localization (datasheet): Membrane ; Multi-pass membrane protein . Melanosome . Identified by mass spectrometry in melanosome fractions from stage I to stage IV.
Tissue details (datasheet): Expressed mostly in brain, muscle, and pancreas but detected in all tissues examined.
Sequence similarities (datasheet): Belongs to the protein kinase superfamily. TKL Ser/Thr protein kinase family. Pelle subfamily.
Research relevance and current trends
- Commonly studied in contexts related to Amino Acid Metabolism,Amino Acids,Channels,Metabolic Signaling Pathways,Metabolism,Pathways and Processes,Plasma Membrane,Signal Transduction.
- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
