Product Overview
This rabbit polyclonal antibody detects suppressor of cytokine signaling 1 (gene SOCS1) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 23.6 kDa, and the supplier reports an observed band at about 27 kDa.
Suppressor of cytokine signaling 1 (SOCS1) is a key negative feedback regulator of interferon and other cytokine signals, including IL-2, IL-4 and IL-6. It binds JAK kinases and receptor chains to block JAK/STAT signaling, and through its SOCS box it also targets proteins for ubiquitin-mediated degradation, so it is widely studied in immune regulation and inflammation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
Samples with a confirmed band (WB): human K562 cells, rat spleen tissue, rat PC-12 cells, mouse spleen tissue, mouse RAW264.7 cells, mouse ANA-1 cells, mouse EL-4 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human cervical cancer tissue, human liver cancer tissue, human esophageal squamous carcinoma tissue, human lung cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the middle region of human SOCS1. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places suppressor of cytokine signaling 1 in the nucleus and cytoplasmic vesicle. Tissue expression noted by UniProt (human): Expressed in all tissues with high expression in spleen, small intestine and peripheral blood leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
